Depends. Who are you saying it to? Older/younger, person you know/don't know, teacher/friend/familiy, etc. I won't arm you with a phrase that might get you punched out.

I'd like to start off by asking, what's your definition of "What's up, dude" that you want said in Japanese?

Are you expecting "Oh, I am doing this and that" or something more like "Hey" in response. I am sure you want something casual and can possible be portrayed as rude depending on the situation. It's just that that little English phrases has so much slang packed into it, it's hard to translate and keep the same attitude and feel.
