No. It has two mistakes.

1. the verb choice
2. the tense

Hold on, let me think. The verb is also wrong, so I guess I cannot say "奥さんは日本で勤めているんですか." ? Maybe I should just stick with ""奥さんは日本で働いてるんですか."

Geeee, mind giving me some examples? :D

Same level of formality as yours:
奥さんは日本で働いてるんですか。
奥さんは日本で仕事をしているんですか。

A tad bit more formal:
Replace the ん by の.

More formal:
~~しているのですか > ~~していらっしゃるので すか

Even more formal:
~~していらっしゃるのですか > ~~してしておら れるのですか

The most formal without being rediculously and/or excessively formal:
~~してしておられるのですか > ~~されておられ るのですか
